At the time of the 2010 Census, Pontotoc had a population of 5,625 people (or 18.78% of the total population of Pontotoc County). This makes Pontotoc the county's most populous community.

The population of Pontotoc represents 0.19% of the population of Mississippi (which has 2,967,297 people), making it the state's 58th most populous community.

With a 2010 count of 5,625 people, the population of Pontotoc increased 7.08% from the 2000 Census (which had a count of 5,253 people).
