At the time of the 2010 Census, Sinai had a population of 120 people. This makes Sinai the county's 7th most populous community.

The population of Sinai represents 0.01% of the total population of South Dakota (which has 814,180 people), making it the state's 291st most populous community.

With a 2010 count of 120 people, the population of Sinai decreased 9.77% from the 2000 Census (with a count of 133 people).
