At the time of the 2010 Census, Saint John had a population of 6,517 people. This makes Saint John the county's 32nd most populous community.

The population of Saint John represents 0.11% of the population of Missouri (which has 5,988,927 people), making it the state's 109th most populous community.

With a 2010 count of 6,517 people, the population of Saint John decreased 5.15% from the 2000 Census (with a count of 6,871 people).
