Taken from the 2019 Census Estimates, Mount Vernon had an population of 4,508 people. This is a decrease of 1.46% since the 2010 Census (or an increase of 12.22% since the 2000 Census).

At the time of the 2010 Census, Mount Vernon had a population of 4,575 people (which is 11.84% of the total population of Lawrence County). This makes Mount Vernon the county's 2nd most populous community.

The population of Mount Vernon represents 0.08% of the total population of Missouri (which has 5,988,927 people), making it the state's 141st most populous community.

With a 2010 count of 4,575 people, the population of Mount Vernon increased 13.89% from the 2000 Census (which had a count of 4,017 people).
