Taken from the 2019 Census Estimates, Jefferson City had an population of 42,708 people. This is a decrease of 0.86% since the 2010 Census (or an increase of 7.75% since the 2000 Census).

At the time of the 2010 Census, Jefferson City had a population of 43,079 people (or 56.69% of the total population of Cole County). This makes Jefferson City the county's most populous community.

The population of Jefferson City represents 0.72% of the total population of Missouri (which has 5,988,927 people), making it the state's 15th most populous community.

With a 2010 count of 43,079 people, the population of Jefferson City increased 8.69% from the 2000 Census (which had a count of 39,636 people).
