Taken from the 2019 Census Estimates, Clayton had an population of 16,747 people. This is an increase of 5.07% since the 2010 Census (or an increase of 30.58% since the 2000 Census).

At the time of the 2010 Census, Clayton had a population of 15,939 people. This makes Clayton the county's 13th most populous community.

The population of Clayton represents 0.27% of the total population of Missouri (which has 5,988,927 people), making it the state's 46th most populous community.

With a 2010 count of 15,939 people, the population of Clayton increased 24.28% from the 2000 Census (which had a count of 12,825 people).
